Die Pfitzner-Moffatt-Oxidation, oft auch einfach Moffat-Oxidation, ist eine milde Oxidationsmethode, die primΓ€re und sekundΓ€re Alkohole mit N,Nβ²-Dicyclohexylcarbodiimid (DCC) und Dimethylsulfoxid (DMSO) in die jeweiligen Aldehyde oder Ketone umwandelt. Speziell hierbei ist, dass die Oxidation bei primΓ€ren Alkoholen nach dem ersten Oxidationsschritt stehen bleibt und nicht zur CarbonsΓ€ure weiter verlΓ€uft, wie dies bei einer herkΓΆmmlichen Behandlung mit Oxidationsmitteln der Fall ist.cite-ref-1[1]cite-ref-2[2]
Vergleichbare Reaktionen sind die Swern-Oxidation (hΓΆhere Ausbeute mit weniger Nebenprodukten), die Dess-Martin-Oxidation, die Julia-Oxidation oder die Oxidation mit Pyridiniumchlorochromat.

Reaktionsmechanismus
Im ersten Schritt wird das Dimethylsulfoxid (DMSO) mit N,Nβ²-Dicyclohexylcarbodiimid (DCC) aktiviert.
Das aktivierte DMSO wird nun von dem Alkohol nukleophil angegriffen. Durch Umlagerungen und Abspaltung entsteht als reaktive Zwischenstufe ein mesomer stabilisiertes Alkoxysulfonium-Ylid. Eine Ξ²-Eliminierung mit zyklischem Γbergangszustand unter Abspaltung von Dimethylsulfid fΓΌhrt zum Endprodukt.
